    • @mutilationisnojoke:

      Good morning,
I am Vincenzo Aiello, President of Foregen.
Foregen is doing well, we have just finished the first set of experiments with mice and sheep, and the results of histology studies tell us we are going in the right direction. The studies with animals show that our decellularized foreskin is biocompatible with mice and is perfectly integrated with the skin of sheep, which is fantastic. We had a meeting with all the scientists and we feel that a second set of experiments with sheep is necessary, We are about to request authorization from the local Ministry of Health. With these new experiments, we intend to perfect the technique of surgery, starting from putting the foreskin vertical, we have to fit the size of each foreskin with each patient, in order to avoid future phimosis, we have to embed the foreskin at the right depth, in the shaft skin, and I feel we have to do it many times before we go to a human patient, we feel a huge responsibility for the first patient and we want to be sure that our procedure is 100% safe before performing it on a human patient.
As you know we are using adult foreskins from cadavers, we have a technology that allows us to remove all the cells from the tissues ( so in future transplants we won't have rejection)and leave the only Extra Cellular Matrix, our goal is to repopulate this matrix with recipient's cells. IF EVERYTHING GOES WELL, in the last part of next year we intend to transplant the "decellularized" foreskins to human patients in a clinical trial. Let's cross our fingers together, and thank you for your extraordinary support.“

      tl,dr:

We are thrilled to announce the completion of the histological analysis of decellularized tissue implanted in sheep by Dr. Angela Palumbo, and the results have exceeded our expectations. This is a significant step towards achieving our mission of creating functional human foreskin tissue.

"Things are super positive," she wrote excitedly to Enzo. "There are many blood vessels well-shaped, and the scaffold is re-epithelialized. It's a fantastic thing."

In simple terms, Dr. Palumbo's findings indicate that the implanted tissue has undergone successful re-epithelialization, and healthy blood vessels have formed in the scaffold. This is a crucial step in regenerating fully functional human foreskin tissue as it indicates that host cells have colonized the tissue. Before attempting to integrate foreskin tissue in human hosts, it is vital to prove that the colonization of a scaffold by host cells is possible. These findings are essential in helping us prepare for human trials.

We want to express our gratitude to you, our dedicated donors, for making this exciting, innovative, and impactful research possible.

      Sincerely,

      Ryan Jones
      Chief Operations Officer

Recently there has been growing interest in regenerative medicine as a means to regenerate the human male foreskin. This option, unlike foreskin restoration, would result in a true human male foreskin being regrown. Pioneers in the field include Dr. Anthony Atala of the Wake Forest Institute of Regenerative Medicine (WFIRM).[15]
In early 2010, Foregen, a non-profit organization dedicated to funding a clinical trial for the purposes of regrowing the human male foreskin, had been founded. If they raise enough funds, a clinical trial has been scheduled for late 2010.[16]
The proposed method would involve placing the patient under general anaesthesia. The penile skin will be opened at the circumcision scar, while the scar tissue is surgically debrided. A biomedical solution will then be applied to both ends of the wound, which will make use of the DNA in the patient's own cells to cause the foreskin to regenerate. A biodegradable scaffold will be used to offer support for the regenerating foreskin.[17]
      2012 gab es ein Update:

...A clinical trial had been scheduled for late 2010, but there were insufficient donations to follow through.[17] The goal was reached in June 2012. The results of the clinical trial will be released in the autumn of 2012.[18]...

      2018:

There has been remarkable success in the field of regenerative medicine in the last two decades. Several functional body parts and organs have been regenerated.

Recently there has been growing interest in regenerative medicine as a means to regenerate the human male foreskin. This option, unlike foreskin restoration, would result in a true human male foreskin being regrown. Pioneers in the field of regenerative medicine include Dr. Anthony Atala of the Wake Forest Institute of Regenerative Medicine (WFIRM).[19] Dr. Atala has been successful in regenerating a functional rabbit penis, but a human penis has not yet been regenerated.

In early 2010, Foregen, an Italian non-profit lobbying organization dedicated to finding interested research groups for the purposes of regrowing the human male foreskin using the regenerative capabilities of the extracellular matrix, in the hopes of eventually being able to provide regeneration to circumcised men, was founded. Unable to do so, Foregen transitioned to a non-profit, dependent upon charitable donations to accomplish its research, with it's own facilities. Foregen is dependent upon charitable donations to accomplish its research. A clinical trial had been scheduled for late 2010, before it transitioned into it's own research organization, but there were insufficient donations to follow through.[20] The goal was reached in June 2012 and they managed to obtain a laboratory and cooperation from biochemists and regenerative medicine experts. Results from their first clinical trial, decellurizing bull foreskins to create the extracellular matrix, were released in February 2013 and as of March 2018, have completed trials on cadaver human foreskins, which was published last year(2017), and is now in the process of peer review[21]. Clinical trials on human subjects might be achieved by 2019.[22]

The proposed method would involve placing the patient under general anaesthesia. The penile skin would be opened at the circumcision scar, while the scar tissue is surgically debrided. A biomedical solution would then be applied to both ends of the wound, causing the foreskin to regenerate with the DNA in the patient's own cells. A biodegradable scaffold (i.e. the de-cellularized foreskin of a cadaver)[23] would be used to offer support for the regenerating foreskin.[24] While social forums have assisted in the discussion of current tools and hurdles towards this method,[25] scientific and medical institutes, as well as bioengineering companies must be involved for a more professional and meaningful discourse and plan of action, including a more reliable method of acquiring cadaver foreskins.

Foreskin regeneration along with laboratory-grown penises (regeneration of the entire penis) could be possible very soon. In 2014, it was announced that it might be possible five years from the initial announcement.[26] It is unclear whether or not having a restored foreskin with full coverage of the glans could interfere with fitting a foreskin scaffold on the penis and subsequent regeneration.[27]

Foreskin regeneration is entirely experimental at this time. It must be emphasized that, as of this writing (March 2018), no human foreskin has yet been regenerated, nor has one been attached to a human. There can be no assurance that regeneration of portions of the human body, including human foreskin regeneration, would be successful in the near future.

We're thrilled to share the significant strides we've made in our sheep trials. Having successfully achieved revascularization and re-epithelialization, we're getting closer to our ultimate goal. So, we've hosted a special September Fundraiser on GoFundMe. Your generous support will help bring us closer to the finish line!

      The Journey So Far

Our initial trials involved attaching decellularized foreskin samples directly onto the backs of sheep. Unfortunately, due to inadequate blood flow, necrosis occurred in several implantations. But every setback is an opportunity for learning. We went back to the drawing board, refined our approach, and embarked on the next round of trials.

In our subsequent trials, we applied foreskin samples to the udder area of sheep. We saw encouraging improvements, with revascularization observed in several sheep. Yet, challenges emerged as the sheep started biting and scratching the area.

      Our Current Progress

For our latest trials, we've innovated an approach that ensures not just revascularization but also re-epithelialization. By burying the foreskin sample tissue beneath a flap of skin on the sheep, the sheep's body itself became a natural bioreactor, creating a vascular network. This method, if successful in humans, could help us avoid necrosis and promote full tissue integration.

Our final vision is a two-stage surgical technique where the decellularized ECM is partially buried beneath the patient's epidermis, then removed and properly attached in the correct position on the penis. To refine this technique and eliminate uncertainties, we need to conduct a final round of experimentation on sheep, set to begin in September 2023.

      Our Ask

We need your help. This crucial final round of experimentation costs approximately $100,000. This will pay for the tissue samples, the sheep, the veterinary specialists, as well as the laboratory space. It's a necessary step before we can move forward to human clinical trials in late 2024. Your support will enable us to conduct these vital experiments, bringing us closer to providing a regenerative solution for circumcised men. Please contribute to our September Fundraiser hosted by GoFundMe!

Your support can offset the costs of this vital phase and propel us forward as we approach the final stage of our research. We're deeply encouraged by the progress we've made, and we're excited about the possibilities that lie ahead. Pledge your support today and join our efforts to be the first in history to fully reverse the harms of male circumcision.

Thank you for standing with us. Together, we may make the difference.

      Sincerely,

      The Foregen Team

$ überschüttet, "Bioscience Company of the Year", 9 Milliarden Dollar schwer - und dann ein riesiger Schwindel.
Alle waren begeistert, alle sind darauf reingefallen: Safeway, Walgreens, George Shultz, William Perry, Henry Kissinger, Sam Nunn..... Die HBO-Doku "The Inventor: Out for Blood in Silicon Valley" ist äußerst sehenswert.
